Yes, you can install a 3500 diff in a 2000 TC.  I did such an installation a
couple of years ago.  You get a stronger diff, one with 4 spider gears, not
just 2, and a beefed up case.  It is a straight forward bolt-in job. The
engine runs at lower revs while cruising but you do pay an acceleration
penalty on takeoff.
